ABSTRACT

The Sit4 protein phosphatase plays a key role in orchestrating various cellular processes essential for maintaining cell viability during aging. We have previously shown that SIT4 deletion promotes vacuolar acidification, mitochondrial derepression, and oxidative stress resistance, increasing yeast chronological lifespan. In this study, we performed a proteomic analysis of isolated vacuoles and yeast genetic interaction analysis to unravel how Sit4 influences vacuolar and mitochondrial function. By employing high-resolution mass spectrometry, we show that sit4Δ vacuolar membranes were enriched in Vps27 and Hse1, two proteins that are part of the endosomal sorting complex required for transport-0. In addition, SIT4 exhibited a negative genetic interaction with VPS27, as sit4∆vps27∆ double mutants had a shortened lifespan compared to sit4∆ and vps27∆ single mutants. Our results also show that Vps27 did not increase sit4∆ lifespan by improving protein trafficking or vacuolar sorting pathways. However, Vps27 was critical for iron homeostasis and mitochondrial function in sit4∆ cells, as sit4∆vps27∆ double mutants exhibited high iron levels and impaired mitochondrial respiration. These findings show, for the first time, cross-talk between Sit4 and Vps27, providing new insights into the mechanisms governing chronological lifespan.

ABSTRACT

Work in the prison system can present a high risk to the mental and physical health of prison officers due to the dangerousness, diversity, and complexity associated with this job. Therefore, knowing that prison officers play a key role in prisons, it is essential to explore variables that help understand the health and well-being of these professionals. Thus, this study aims to explore and describe the relationship between social support, generalized anxiety, and life satisfaction in a sample of 100 Portuguese prison officers. The results showed the existence of a significant positive relationship between social support and life satisfaction. Social support also shows a significant negative relationship, decreasing anxiety. It was also found that increased anxiety significantly contributed to a decrease in the participants’ satisfaction with life. It was also possible to verify the existence of a mediating relationship between social support and satisfaction with life through anxiety. The results are subsequently discussed and translated into implications for research and professional practice. (AU)


El trabajo en el sistema penitenciario puede presentar un alto riesgo para la salud mental y física de los funcionarios de prisiones debido a la peligrosidad, diversidad y complejidad asociadas a este trabajo. Por lo tanto, sabiendo que los agentes penitenciarios desempeñan un papel clave en las prisiones, es esencial explorar variables que ayuden a comprender la salud y el bienestar de estos profesionales. Así, este estudio tiene como objetivo explorar y describir la relación entre el apoyo social, la ansiedad generalizada y la satisfacción con la vida en una muestra de 100 agentes penitenciarios portugueses. Los resultados mostraron la existencia de una relación positiva significativa entre el apoyo social y la satisfacción con la vida. El apoyo social también muestra una relación negativa significativa, disminuyendo la ansiedad. También se comprobó que el aumento de la ansiedad contribuía significativamente a la disminución de la satisfacción con la vida de los participantes. También se pudo comprobar la existencia de una relación mediadora entre el apoyo social y la satisfacción con la vida a través de la ansiedad. Posteriormente, se discuten los resultados y se traducen en implicaciones para la investigación y la práctica profesional. (AU)

ABSTRACT

PURPOSE: This study aimed to cross-validate a recently proposed equation for the prediction of maximal oxygen uptake (V˙O2max) in cycling exercise by using the average power output normalized by the body mass from a 5-minute time trial (RPO5-min) as the independent variable. Further, the study aimed to update the predictive equation using Bayesian informative prior distributions and meta-analysis. METHODS: On different days, 49 male cyclists performed an incremental graded exercise test until exhaustion and a 5-minute time trial on a stationary cycle ergometer. We compared the actual V˙O2max with the predicted value obtained from the RPO5-min, using a modified Bayesian Bland-Altman agreement analysis. In addition, this study updated the data on the linear regression between V˙O2max and RPO5-min, by incorporating information from a previous study as a Bayesian informative prior distribution or via meta-analysis. RESULTS: On average, the predicted V˙O2max using RPO5-min underestimated the actual V˙O2max by -6.6 mL·kg-1·min-1 (95% credible interval, -8.6 to -4.7 mL·kg-1·min-1). The lower and upper 95% limits of agreement were -17.2 (-22.7 to -12.3) and 3.8 (-1.0 to 9.5) mL·kg-1·min-1, respectively. When the current study's data were analyzed using the previously published data as a Bayesian informative prior distribution, the accuracy of predicting sample means was found to be better when compared with the data combined via meta-analyses. CONCLUSIONS: The proposed equation presented systematic bias in our sample, in which the prediction underestimated the actual V˙O2max. We provide an updated equation using the previous one as the prior distribution, which could be generalized to a greater audience of cyclists.

ABSTRACT

This study explores the intricate relationship between the yeast vacuolar H+-ATPase (V-ATPase) and neutral lipid metabolism. We show that LD generation observed upon loss of V-ATPase activity is crucial for survival in lipotoxic conditions. Moreover, the study uncovers a link between V-ATPase function, inositol metabolism and the activation of the oxidative pentose phosphate pathway, highlighting its pivotal role in counteracting oxidative stress. This work provides foundational insights into metabolic adaptations triggered by V-ATPase dysfunction, shedding light on cellular adaptability under lipotoxic and oxidative stress conditions.

ABSTRACT

The mitochondrial outer membrane protein porin 1 (Por1), the yeast orthologue of mammalian voltage-dependent anion channel (VDAC), is the major permeability pathway for the flux of metabolites and ions between cytosol and mitochondria. In yeast, several Por1 phosphorylation sites have been identified. Protein phosphorylation is a major modification regulating a variety of biological activities, but the potential biological roles of Por1 phosphorylation remains unaddressed. In this work, we analysed 10 experimentally observed phosphorylation sites in yeast Por1 using bioinformatics tools. Two of the residues, T100 and S133, predicted to reduce and increase pore permeability, respectively, were validated using biological assays. In accordance, Por1T100D reduced mitochondrial respiration, while Por1S133E phosphomimetic mutant increased it. Por1T100A expression also improved respiratory growth, while Por1S133A caused defects in all growth conditions tested, notably in fermenting media. In conclusion, we found phosphorylation has the potential to modulate Por1, causing a marked effect on mitochondrial function. It can also impact on cell morphology and growth both in respiratory and, unpredictably, also in fermenting conditions, expanding our knowledge on the role of Por1 in cell physiology.

ABSTRACT

A critical step to design wildlife mitigating measures is the identification of roadkill hotspots. However, the effectiveness of mitigations based on roadkill hotspots depends on whether spatial aggregations are recurrent over time, spatially restricted, and most importantly, shared by species with diverse ecological and functional characteristics. We used a functional group approach to map roadkill hotspots for mammalian species along the BR-101/North RJ, a major road crossing important remnants of the Brazilian Atlantic Forest. We tested if functional groups present distinct hotspot patterns, and if they converge into the same road sectors, in that case, favoring optimal mitigating actions. Roadkill rates were monitored and recorded between October/2014 and September/2018 and species were classified into six functional groups based on their home range, body size, locomotion mode, diet, and forest-dependency. Hotspots along the roads were mapped for comparison of spatial patterns between functional groups. Results demonstrated that the roadkill index varied idiosyncratically for each functional group throughout the months and that no group presented seasonality. Seven hotspots were shared by two or more functional groups, highlighting the importance of these road stretches to regional mammal fauna. Two of the stretches are associated with aquatic areas extending from one side of the road to the other, and the remaining are connected to patches of native vegetation on both sides. This work brings a promising approach, yet hardly used in ecological studies on roads to analyze roadkill dynamics, assigning more importance to ecological instead of taxonomical characteristics, normally used to identify spatiotemporal patterns.

ABSTRACT

Despite the considerable advancements in oncology, cancer remains one of the leading causes of death worldwide. Drug resistance mechanisms acquired by cancer cells and inefficient drug delivery limit the therapeutic efficacy of available chemotherapeutics drugs. However, studies have demonstrated that nano-drug carriers (NDCs) can overcome these limitations. In this sense, exosomes emerge as potential candidates for NDCs. This is because exosomes have better organotropism, homing capacity, cellular uptake, and cargo release ability than synthetic NDCs. In addition, exosomes can serve as NDCs for both hydrophilic and hydrophobic chemotherapeutic drugs. Thus, this review aimed to summarize the latest advances in cell-free therapy, describing how the exosomes can contribute to each step of the carcinogenesis process and discussing how these nanosized vesicles could be explored as nano-drug carriers for chemotherapeutics.

ABSTRACT

As centers for energy production and essential biosynthetic activities, mitochondria are vital for cell growth and proliferation. Accumulating evidence suggests an integrated regulation of these organelles and the nuclear cell cycle in distinct organisms. In budding yeast, a well-established example of this coregulation is the coordinated movement and positional control of mitochondria during the different phases of the cell cycle. The molecular determinants involved in the inheritance of the fittest mitochondria by the bud also seem to be cell cycle-regulated. In turn, loss of mtDNA or defects in mitochondrial structure or inheritance often lead to a cell cycle delay or arrest, indicating that mitochondrial function can also regulate cell cycle progression, possibly through the activation of cell cycle checkpoints. The up-regulation of mitochondrial respiration at G2/M, presumably to fulfil energetic requirements for progression at this phase, also supports a mitochondria-cell cycle interplay. Cell cycle-linked mitochondrial regulation is accomplished at the transcription level and through post-translational modifications, predominantly protein phosphorylation. Here, we address mitochondria-cell cycle interactions in the yeast Saccharomyces cerevisiae and discuss future challenges in the field.

ABSTRACT

Niemann-Pick type C1 (NPC1) is an endolysosomal transmembrane protein involved in the export of cholesterol and sphingolipids to other cellular compartments such as the endoplasmic reticulum and plasma membrane. NPC1 loss of function is the major cause of NPC disease, a rare lysosomal storage disorder characterized by an abnormal accumulation of lipids in the late endosomal/lysosomal network, mitochondrial dysfunction, and impaired autophagy. NPC phenotypes are conserved in yeast lacking Ncr1, an orthologue of human NPC1, leading to premature aging. Herein, we performed a phosphoproteomic analysis to investigate the effect of Ncr1 loss on cellular functions mediated by the yeast lysosome-like vacuoles. Our results revealed changes in vacuolar membrane proteins that are associated mostly with vesicle biology (fusion, transport, organization), autophagy, and ion homeostasis, including iron, manganese, and calcium. Consistently, the cytoplasm to vacuole targeting (Cvt) pathway was increased in ncr1∆ cells and autophagy was compromised despite TORC1 inhibition. Moreover, ncr1∆ cells exhibited iron overload mediated by the low-iron sensing transcription factor Aft1. Iron deprivation restored the autophagic flux of ncr1∆ cells and increased its chronological lifespan and oxidative stress resistance. These results implicate iron overload on autophagy impairment, oxidative stress sensitivity, and cell death in the yeast model of NPC1.

ABSTRACT

Cdh1p is one of the two substrate adaptor proteins of the anaphase promoting complex/cyclosome (APC/C), a ubiquitin ligase that regulates proteolysis during cell cycle. In this work, using a proteomic approach, we found 135 mitochondrial proteins whose abundance was significantly altered in the cdh1Δ mutant, with 43 up-regulated proteins and 92 down-regulated proteins. The group of significantly up-regulated proteins included subunits of the mitochondrial respiratory chain, enzymes from the tricarboxylic acid cycle and regulators of mitochondrial organization, suggesting a metabolic remodelling towards an increase in mitochondrial respiration. In accordance, mitochondrial oxygen consumption and Cytochrome c oxidase activity increased in Cdh1p-deficient cells. These effects seem to be mediated by the transcriptional activator Yap1p, a major regulator of the yeast oxidative stress response. YAP1 deletion suppressed the increased Cyc1p levels and mitochondrial respiration in cdh1Δ cells. In agreement, Yap1p is transcriptionally more active in cdh1Δ cells and responsible for the higher oxidative stress tolerance of cdh1Δ mutant cells. Overall, our results unveil a new role for APC/C-Cdh1p in the regulation of the mitochondrial metabolic remodelling through Yap1p activity.

Este artigo buscou compreender como a prática esportiva vivenciada no programa Curumim influenciou a relação dos antigos participantes do programa no Sesc Ribeirão Preto com o esporte. Como metodologia, utilizamos a Teoria Fundamentada; seis adultos que participaram do Curumim foram entrevistados. Durante a análise, cinco categorias foram estabelecidas, sendo a principal "Curumim como 'uma janela que se abre': descobertas, aprendizados e influências". Por fim, foi possível evidenciar a influência do Curumim no envolvimento com a prática esportiva. Contudo, este não foi um fator decisivo. Ainda, experiências marcantes demonstraram potencial para alterar o modo de ação de seus frequentadores e definir a escolha profissional dos participantes.


This article sought to understand how the sports practice experienced in the Curumim Program influenced the relationship of the former participants with sport. As methodology, the Grounded Theory was used; six adults who participated in the Curumim in Sesc Ribeirão Preto were interviwed. Analyzing the data, five categories were established and the main one was "Curumim as 'an open window': discoveries, learning and influences". Finally, it was possible to perceive the influence of Curumim in sports practice, however, this was not a decisive factor. Still, remarkable experiences showed the potential to change the way of action of its participants and to define the professional choice.

ABSTRACT

Cdc28, the homolog of mammalian Cdk1, is a conserved key regulatory kinase for all major cell cycle transitions in yeast. We have found that defects in mitochondrial respiration (including deletion of ATP2, an ATP synthase subunit) inhibit growth of cells carrying a degron allele of Cdc28 (cdc28td) or Cdc28 temperature-sensitive mutations (cdc28-1 and cdc28-1N) at semi-permissive temperatures. Loss of cell proliferation in the atp2Δcdc28td double mutant is associated with aggravated cell cycle arrest and mitochondrial dysfunction, including mitochondrial hyperpolarization and fragmentation. Unexpectedly, in mutants defective in mitochondrial respiration, steady-state protein levels of mutant cdc28 are strongly reduced, accounting for the aggravated growth defects. Stability of Cdc28 is promoted by the Hsp90-Cdc37 chaperone complex. Our results show that atp2Δcdc28td double-mutant cells, but not single mutants, are sensitive to chemical inhibition of the Hsp90-Cdc37 complex, and exhibit reduced levels of additional Hsp90-Cdc37 client kinases, suggesting an inhibition of this complex. In agreement, overexpression of CDC37 improved atp2Δcdc28td cell growth and Cdc28 levels. Overall, our study shows that simultaneous disturbance of mitochondrial respiration and Cdc28 activity reduces the capacity of Cdc37 to chaperone client kinases, leading to growth arrest.

ABSTRACT

O estudo tem como objetivo descrever os comportamentos de proteção adotados por estudantes portugue-ses do ensino superior durante a pandemia e analisar a sua relação com a percepção de risco e o papel mediador do medo face à covid-19. Participaram 335 estu-dantes com idades entre os 18 e os 29 anos (m= 21.42; dp= 2.43). Os participantes preencheram um inquérito sobre os comportamentos de proteção (i.e., preventi-vos e de evitamento), a percepção de risco e o medo face à covid-19. Os resultados demonstraram que os comportamentos preventivos mais utilizados foram a lavagem/desinfecção das mãos ao longo do dia e o uso de máscara na via pública ou espaços exteriores. Os comportamentos de evitamento mais adotados foram o de evitar locais com aglomeração e os convívios presenciais com colegas/amigos. Verificouse uma as-sociação positiva entre os comportamentos de proteção, a percepção de gravidade e o medo face à covid-19. O medo face à covid-19 foi mediador da relação entre a percepção de gravidade e os comportamentos preventivos (c' = .26, ic 95% [.11; .44]) e de evitamento (c' = .28, ic 95% [.12; .50]). Os resultados são discutidos aten-dendo ao papel da percepção de risco e do medo nas respostas comportamentais dos jovens, num contexto de pandemia, sendo apresentadas implicações práticas e sugestões para estudos futuros.


El estudio tiene como objetivo describir las conductas protectoras adoptadas por los estudiantes portugueses de educación superior durante la pandemia, y analizar su relación con la percepción de riesgo y el papel mediador del miedo al covid-19. Participaron en el estudio 335 estudiantes de entre 18 y 29 años (m = 21.42; ds = 2.43). Los participantes completaron una encuesta sobre conductas protectoras (es decir, preventivas y de evitación), percepción de riesgo y miedo al covid-19. Los resultados mostraron que las conductas preventivas más utilizadas fueron el lavado/desinfección de manos a lo largo del día y el uso de mascarilla en la vía pública o al aire libre. Los comportamientos de evitación más adoptados fueron evadir lugares con reuniones e interacciones cara a cara con compañeros/amigos. Hubo una correlación positiva entre las conductas protectoras, la percepción de seriedad y el miedo al covid-19. También se encontró que el miedo al covid-19 medió la relación entre la percepción de severidad y las conductas preventivas (c' = .26, ic 95% [.11; .44]) y de evitación (c' = .28, ic 95% [.12; .50]). Los resultados se discuten considerando el papel de la percepción del riesgo y el miedo en las respuestas conductuales de los jóvenes en un contexto pandémico, así mismo se presentan impli-caciones prácticas y sugerencias para estudios futuros.


The study aims to describe the protective behaviors adopted by Portuguese higher education students in during the and to analyze their relationship with the perception of risk and the mediating role of fear of covid-19. A total of 335 students aged be-tween 18 and 29 participated in the study (m= 21.42; sd = 2.43). Participants completed a survey on protective (i.e., preventive and avoidance) behaviors, risk perception, and fear of covid-19. The results showed that the most used preventive behaviors were hand washing/disinfection throughout the day and using masks in public streets or outdoor spaces. The most adopted avoidance behaviors were avoiding crowded places and face-to-face interactions with colleagues/friends. There was a positive correlation between pro-tective behaviors, the perception of severity, and fear of covid-19. Fear of covid-19 mediated the relation between the perception of severity and both preventive (c' = .26, ic 95% [.11; .44]) and avoidance (c' = .28, ic95% [.12; .50]) behaviors. The results are discussed considering the role of risk perception and fear in the behavioral responses of young people in a pandemic context while presenting practical implications and suggestions for future studies.

ABSTRACT

Higher education students have faced several changes in their lives due to the COVID-19 pandemic. This study aims to explore the effect of dispositional optimism in students' fear of COVID-19 and to test the mediating role of general anxiety in the relationship between optimism and fear. Using an online survey, data were collected during the second wave of the pandemic in Portugal. The sample included 312 higher education students (76% females) aged 18-25 years old, who completed measures of dispositional optimism, general anxiety and fear of COVID-19. The results showed that higher optimism and lower general anxiety reduce fear of COVID-19. Moreover, the link between optimism and fear is fully mediated by general anxiety, showing that optimism reduces fear of COVID-19 indirectly through the reduction of students' anxiety. The role of optimism, anxiety and fear in higher education students is discussed and topics for further research are presented.

ABSTRACT

The objective of this study was to evaluate the influence of the extraction socket (distal or lingual root) and the type of region of interest (ROI) definition (manual or predefined) on the assessment of alveolar repair following tooth extraction using micro-computed tomography (micro-CT). The software package used for scanning, reconstruction, reorientation, and analysis of images (NRecon®, DataViewer®, CT-Analyzer®) was acquired through Bruker < https://www.bruker.com > . The sample comprised the micro-CT volumes of seven Wistar rat mandibles, in which the right first molar was extracted. The reconstructed images were analyzed using the extraction sockets, i.e., the distal and intermediate lingual root and the method of ROI definition: manual (MA), central round (CR), and peripheral round (PR). The bone volume fraction (BV/TV) values obtained were analyzed by two-way ANOVA with Tukey's post hoc test (α = 5%). The distal extraction socket resulted in significantly lower BV/TV values than the intermediate lingual socket for MA (P = 0.001), CR (P < 0.001), and PR (P < 0.001). Regarding the ROI, when evaluating the distal extraction socket, the BV/TV was significantly higher (P < 0.001) for MA than for CR and PR, with a lower BV/TV for CR. However, no significant difference was observed for MA (P = 0.855), CR (P = 0.769), or PR (P = 0.453) in the intermediate lingual extraction socket. The bone neoformation outcome (BV/TV) for alveolar bone repair after tooth extraction is significantly influenced by the ROI and the extraction socket. Using the predefined method with a standardized ROI in the central region of the distal extraction socket resulted in the assessment of bone volume, demonstrating the most critical region of the bone neoformation process.

ABSTRACT

Despite the considerable advancements in oncology, cancer remains one of the leading causes of death worldwide. Drug resistance mechanisms acquired by cancer cells and inefficient drug delivery limit the therapeutic efficacy of available chemotherapeutics drugs. However, studies have demonstrated that nano-drug carriers (NDCs) can overcome these limitations. In this sense, exosomes emerge as potential candidates for NDCs. This is because exosomes have better organotropism, homing capacity, cellular uptake, and cargo release ability than synthetic NDCs. In addition, exosomes can serve as NDCs for both hydrophilic and hydrophobic chemotherapeutic drugs. Thus, this review aimed to summarize the latest advances in cell-free therapy, describing how the exosomes can contribute to each step of the carcinogenesis process and discussing how these nanosized vesicles could be explored as nano-drug carriers for chemotherapeutics.

ABSTRACT: Borszcz, FK, Vieira, MT, Tramontin, AF, Visentainer, LH, and Costa, VP. Is functional overreaching or acute fatigue the key to the effects of concentrated block training in running? J Strength Cond Res 36(12): 3485-3496, 2022-This study examined the effects of 5 consecutive days of high- and moderate-intensity training on performance and physiological measures in moderately trained individuals. The relationship of the training organization with the state of overreaching and acute fatigue was investigated. Twenty-four male soldiers (age, 19.3 ± 0.4 years; V̇o2peak, 58.7 ± 3.8 ml·kg-1·min-1) were assigned to 2 training groups for 5 consecutive days of either high- or moderate-intensity training. The subjects underwent incremental and 12-minute time trial tests before, immediately after, 1 and 2 weeks after training. The high- and moderate-intensity sessions were 30 minutes in duration and performed at fixed velocities of 13.3 and 10 km·h-1 (near second and first ventilatory thresholds), respectively. Acute fatigue and overreaching criteria were set as concomitant nonimpairment and impairment, respectively, in the incremental peak velocity and 12-minute time trial performances at posttest immediately after the training block. Data analyses were completed using hierarchical Bayesian's models. In subjects who wer trained at moderate intensity, no performance impairment occurred (i.e., acute fatigue); for the high-intensity training, 5 subjects showed impairment in performance and were classified as overreached. Only in subjects who were acutely fatigued, clear beneficial effects were observed in incremental test peak velocity and 12-minute time trial performances. In moderately trained runners, a block of 5 consecutive days of moderate-intensity training was demonstrated to be a useful strategy for the improvement of performance. However, high-intensity training does not seem to be a safe strategy because of the observations of overreaching and inferior probabilities of performance improvements.

ABSTRACT

INTRODUCTION: Pediatric acute myeloid leukemia (AML) is the second most common type of pediatric leukemia. Patients with AML are at high risk for several complications such as infections, typhlitis, and acute and long-term cardiotoxicity. Despite this knowledge, there are no definite supportive care guidelines as to what the best approach is to manage or prevent these complications. AREA COVERED: The NOPHO-DB-SHIP (Nordic-Dutch-Belgian-Spain-Hong-Kong-Israel-Portugal) consortium, in preparation for a new trial in pediatric AML patients, had dedicated meetings for supportive care. In this review, the authors discuss the available data and outline recommendations for the management of children and adolescents with AML with an emphasis on hyperleukocytosis, tumor lysis syndrome, coagulation abnormalities and bleeding, infection, typhlitis, malnutrition, cardiotoxicity, and fertility preservation. EXPERT OPINION: Improved supportive care has significantly contributed to increased cure rates. Recommendations on supportive care are an essential part of treatment for this highly susceptible population and will further improve their outcome.
